Pre Treatment Note:1. Before blowing down eluent add 300µL of 0.5N methanolic hydrochloride (to prevent loss of analyte) 2. Dilute sample by a factor of 20 before injecting onto LCMS (Reconstitute in 10% methanol in formic acid (0.1%)).
SPE Load:Load 4.00 mL Sample diluted in 100mM Phosphate buffer
SPE Wash 1:2mL of 100mM sodium acetate buffer pH 5.0
SPE Wash 2:2mL of Methanol
SPE Dry:10 min at 10 inches of Hg
SPE Elution 1:2mL of Ethyl acetate: IPA:NH4OH (7:2:1)
SPE Evaporation:Evaporate to dryness
SPE Reconstitute:1000 µL 10% methanol in formic acid (0.1%)
